Long-term StoragePod / MultiPod performance data from FIMM

FIMM HTB (High Throughput Biomedicine) laboratory in Helsinki

StoragePod performance data is kindly provided by FIMM HTB Unit in Helsinki . FIMM focuses on personalized medicine and human genomics. The primary function of the HTB unit is to provide research services related to high-throughput screening approaches both with biochemical and cell based assays. HTB is providing support and performing overall project planning, assay development, assay miniaturization, screening, running high quality-controlled follow-up assays and validations and data analysis and interpretation. Also, HTB is providing bioinformatics tools for analyzingdrug screening data with single drugs or with drug combinations.  HTB is offering users “assay ready” plates or tubes with bioactive and drug-like chemicals or siRNAs. This covers multiple sites across Europe and world wide.

FIMM HTB has been a StoragePod user since 2010. They have a MultiPod Controller with 4 San Francisco StoragePods and 70 Dundee StoragePods. ECHO dispensers (3) are used for liquid handling. StoragePods are used to both store plates/compounds and to also periodically recover plates by removing moisture absorbed into their DMSO. StoragePods are stored at -20°C, +4°C and all frequently used compound stocks are in StoragePods at room temperature. HTB runs regular functional QC checks to their compound collections and have kindly provided QC data for their FO5, 5th generation oncology collection. HTB have developed a Drug Sensitivity Score (DSS) that is based on multiple parameters. It is robust to various experimental settings, and enables compound activity and response comparison.

Functional QC: Trendline of Compound X activity over the years

We know Compound X is losing activity when exposed to air and moisture on plate, but the compound dissolved in DMSO remains active when stock tube is stored under nitrogen gas.

There is usually some variation from experiment to experiment depending on the compound and the cell line, but here also compound stability on the ECHO source plate is causing variation. [What is being seen in the chart above is plate to plate variance, with all of the plates being made from the same tube. The DSS is narrow in a 17-24 range.]

Stock has been dissolved in DMSO since 2015.

Plots are consistent over 8 years – any differences are from assay-assay variation.

 

Summary

  • The results/DSS show that the compounds are being well maintained over an extended period, with relatively consist DSS and % inhibition.
  • This shows that the Roylan StoragePods provide a good system for compound storage. And, also for the recovery of plates by dehydrating them free of moisture. To maintain high DMSO purity and thus good compound solubility and concentration levels.
  • FIMM HTB has been supplying compound plates to its customers/collaborators for many years with consistent performance of the supplied compounds.
